This c ontamination presents appreciable\ndisposal challenges and environmental r isks.  This work explores the\nmechanism of elemental release to fluid as well as novel methods of\nretaining some elements in the solid phase. Ele ments explored in depth\ninclude arsenic\, uranium\, and rare earth elemen ts.  Experiments\nindicate that organic ligands such as bipyridine are ca pable of\nsequestering >95% of uranium in novel solid phases\, and less to xic\nligands such as histidine are also somewhat effective.  Inorganic\ns orption of metals on hydroxyapatite and formation of\nuranium-phosphate mi nerals is also observed.  The applicability of\nthese methods to fracking conditions is explored.\n\nBIOGRAPHY: Alison Keimowitz is an Associate Pr ofessor of Chemistry at\nVassar College\, and also serves as the director of the Environmental\nStudies program. Bassler.\n\nABOUT THE LECTURE\n\nBacteria communica te with one another via the production and detection\nof secreted signal m olecules called autoinducers. This cell-to-cell\ncommunication process\, c alled “Quorum Sensing”\, allows bacteria to\nsynchronize behavior on a population-wide scale. Behaviors controlled\nby quorum sensing are ones t hat are unproductive when undertaken by an\nindividual bacterium acting al one but become effective when undertaken\nin unison by the group. For exam ple\, quorum sensing controls virulence\nfactor production and biofilm for mation. Bassler’s group discovered\nthat higher organisms that harbor qu orum-sensing bacteria in their\nmicrobiomes participate in these chemical conversations by providing\nthe biological building blocks bacteria need t o make autoinducers.\nBassler’s group also showed that quorum-sensing au toinducer\ninformation can be hijacked by viruses that infect and kill bac teria.\nThe viral strategy of monitoring quorum-sensing autoinducer\naccum ulation and killing the bacterial host exclusively at high cell\ndensity m aximizes viral dissemination to the next host cell. Thus\,\ninteractions a cross the eukaryotic\, bacterial\, and viral domains all\nrely on quorum s ensing. Bassler’s group invented quorum-sensing\ndisruption strategies f or development into new anti-microbials. If IDPs are\nsomewhat like a c ooked noodle\, then condensates are roughly akin to a\nball of cooked spag hetti\, or perhaps pasta primavera. In this talk\, I\nwill describe three recent studies from my lab and collaborators\,\nrelating to the biophysics and bioengineering of these “noodles.”\nI will begin by discussing en gineered IDPs (including highly charged\nsequences) that have provided new insights into the molecular grammar\nof protein phase separation. Second\ , I will present biophysical\ninsights into the role of protein condensati on in the SARS-CoV-2 viral\nlifecycle\, focusing on how phosphorylation wi thin a cationic\ndisordered region toggles the material state and function of\nnucleocapsid protein condensates.
